RESEARCH ACTIVITIES

Applied Basic Research: Focus on the role of endothelium-derived mediators (i.e. nitric oxide, reactive oxygen species, prostaglandin H2, endothelin-1 and tissue factor) in the regulation of vascular tone, platelet-vessel wall interaction, coagulation (tissue factor) as well as vascular structure in physiology and in hypertension, in high LDL-cholesterol and HDL dysfunction and atherosclerosis (with a focus on LOX-1) using cellular and molecular techniques as well as ge-netically engineered animal models. More recently, aging (p66shc) and longevity genes (Sirtuins, jun-D) in wild-type and diabetic mice have been studied.

  Clinical Research: Translation of basic findings and focus on vascular dysfunction in hyper-tension, hyperlipidemia, coronary artery disease and congestive heart failure employing ple-thysmography, ultrasound, angiography, IVUS, optical coherence tomography, positron emis-sion tomography and magnetic resonance imaging. Also, nutritional and pharmacological inter-ventions and of devices and catheter-based interventions are investigated.
  Registries: The natural history has and is further investigated in large registries enrolling pa-tients with acute coronary syndromes (Swiss SPUM–ACS registry; n=4’947; the AMIS-Plus registry , n =50’000, the HEART-ACS registry, Harefield Hospital, n= 600, but still recruiting,), Tako Tsubo Registry (InterTAK Registry; n=2’100; 2014- 2018).
 

Clinical Multicenter Trials: Translation of knowledge into clinical practice in the following trials (all published in high impact journals; see list of publications):

  • ENCORE-1: Effects of a calcium antagonist and/or statin on coronary vasomotion.
  • ENCORE–2: Effects of a calcium antagonsit and/or statin on coronary plaque volume.
  • SIRTAX: Sirolimus- vs. Paclitaxel-coated stents on outcomes in chronic coronary
  • COMFORTABLE: Effect of biolimus-eluting stents with biodegradable polymer vs bare-metal stents on CV events among patients with acute myocardial infarction.

  • EARTH: Effects of of the ETA-antagonist Darusentan on left ventricular remodelling.
  • FAIR-HF: Effectos of iron supplementation on QoL and performance in HFrEF.
  • Dal-VESSEL: Effects of the CETP inhibitor Dalcetrapib on endothelial function. Exetimibe
